چکیده

UNLABELLED A 56-year-old woman with a history of disposable soft contact lens wear was referred to our university eye center for a corneal ulcer. Based on the microbial culture, the initial diagnosis was bacterial keratitis, which was unresponsive to topical fortified antibiotics. The patient was then examined using in vivo confocal microscopy, which revealed Acanthamoeba infection. This case emphasizes the need to suspect Acanthamoeba infection in soft contact lens wearers who present with progressive ulcerative keratitis or progressively worsening corneal ulcers that are not responsive to the usual antimicrobial therapy. It is also important to consider the possibility of a coinfection with bacterial and Acanthamoeba species. Unusual case of Acanthamoeba polyphaga and Pseudomonas aeruginosa keratitis in a contact lens wearer from Gauteng, South Africa.

Acanthamoeba species can cause a chronic, progressive ulcerative keratitis of the eye which is not responsive to the usual antimicrobial therapy and is frequently mistaken for stromal herpes keratitis. An unusual case of coinfection with Acanthamoeba polyphaga and Pseudomonas aeruginosa as causes of corneal keratitis in a contact lens wearer from Gauteng, South Africa, is reported. These two pa...

Activity of the amidoamine myristamidopropyl dimethylamine against keratitis pathogens.

OBJECTIVES Microbial keratitis accounts for up to 30% of blindness in some less developed societies. The development of a single broad-spectrum topical antimicrobial effective against bacteria, fungi and Acanthamoeba would have a major impact on reducing the morbidity and simplifying the treatment of microbial keratitis. To this end, the activity of the amidoamine myristamidopropyl dimethylamin...

Acanthamoeba binds to extracellular matrix proteins in vitro.

PURPOSE To identify host-tissue amoeba interactions that may be important in the pathogenesis of Acanthamoeba keratitis, the ability of the opportunistic pathogen Acanthamoeba polyphaga to bind various components of the extracellular matrix (collagen type IV, laminin, or fibronectin) was examined in vitro. METHODS A polyphaga, isolated from a case of human amoebic keratitis, was used in the s...

Development of colorimetric microtiter plate assay for assessment of antimicrobials against Acanthamoeba.

We have developed and optimized a 96-well microtiter plate assay, based on the reduction of alamarBlue, to assess the efficacies of much needed new antimicrobials against Acanthamoeba species. This assay has been optimized for determination of drug efficacy against two potentially pathogenic species, Acanthamoeba castellanii and Acanthamoeba polyphaga, and has been validated by comparison of th...

Experimental Acanthamoeba keratitis: I. Preliminary findings.

Experimental Acanthamoeba keratitis was induced in Wistar rats by intrastromal inoculation of Acanthamoeba polyphaga cysts. Keratitis developed in all corneas injected and was characterised by a diffuse granular opacity. The animals were examined for 70 days and a gradual reduction of keratitis was observed. Pathological sections showed liquefactive stromal necrosis and Acanthamoeba localised i...
